Genetics at Toronto University.

Dear Professor V

My name is Mahd Rauf, a graduate student, currently studying virology major at National

University of Science and Technology, Pakistan. I am almost finishing my Master of Science

(MS) degree at the same institution and would wish to apply for a Ph.D. in Molecular Genetics at

the University of Toronto, and most importantly to join the group you supervise. I therefore

decided to send this email as an introduction.

I have been tracking your laboratory’s webpage for quite some time and I have really

been fascinated by the work you did on viral proteomics. I would therefore love to join you in

this line of work as your graduate student. I also read your article, “Contributions of the Epstein-

Barr Virus EBNA1 Protein to Gastric Carcinoma,” a very instructive paper, I must say, I was

especially fascinated by your finding that EBNA1 protein interferes with p53 activation and

apoptosis.

I have a good research experience in the field of Molecular Virology. I worked on Phage

Therapy as my undergraduate research. My supervisor, Dr. Shafiq-ur-Rehman, and I were able to

discover a broad-spectrum bacteriophage that lysed several pathogenic bacteria and yet did not

infect any probiotics included in the study. Our study was published in Journal of Basic

Microbiology. I also participated in another virology-based project in my undergraduate school

in which I was to amplify and clone NS5B gene of hepatitis C virus. The gene encodes for RNA

dependent RNA polymerase and is a target of many antivirals. I learnt several techniques during

that short project such as RNA extraction, cDNA synthesis, nested PCR, cloning, transformation,

and western blotting. Finally, my MS research revolved around the anticancer efficacy of a few

novel derivatives of isatin. Though my part in the project was in vitro based, I also learnt how to

make rat models of cirrhosis and hepatocellular carcinoma

I have also studied several subjects related to virology, including molecular virology,

viral pathogenesis, and medical virology. I also wrote a report on the first outbreak of

chikungunya in Pakistan. The report summarised the environmental as well as the anthropogenic

factors behind the outbreak. It was published in the journal, “The Lancet Infectious Diseases.” I

have also designed courses on emerging viral infections and tumor virology for my former

supervisor.

I am sure that my academic and research experiences have impacted me enough to be a

productive member of your research team. I hope you do not mind that I have indicated my

interest in joining your group on my statement of purpose in my application to further my studies

in your institution and department. I would be grateful if you grant my request and become my

I have had sufficient research experience in the field of cancer biology. My Master of

Science research was pillared around hepatocellular carcinoma. I had to assess the anticancer

efficacy of a few novel derivatives. To begin with, I checked for cytotoxic effects of the

compounds on HepG2 cell line. To do so, I treated HepG2 cells with different concentrations of

the compounds and then performed PCR on GAPDH, a housekeeping gene. None of the

compounds interfered with the level of GAPDH transcription. I then checked the anti-

proliferative effects of the compounds on several liver cell lines. For this purpose, I conducted

the following cell-based essays; lactate dehydrogenase, neutral red, trypan blue, XTT, and

propidium iodide. One of the derivatives exhibited significant anti-proliferative effects. I have

also established rat modes of cirrhosis and hepatocellular carcinoma and plan on extending my

study to vivo level.

I am confident that my academic and research experiences have prepared me to be a

My supervisor, Dr. Shafiq-ur-Rehman, and were able to discover a broad-spectrum

bacteriophage that lysed several pathogenic bacteria, yet did not infect any probiotics included in

the study. During the project, I served the following roles: First, I assessed the host range of the

bacteriophage and then checked its bactericidal activity against Pseudomonas and several species

of Enterobacteriaceae. The strain that was included in the study was either extreme or total drug

resistant, and the phage lysed most of them. I then molecularly characterized the bacteriophage.

Its electron microscopy and restriction digestion revealed that it was a member of the family

Siphoviridae. Finally, I assessed the antimicrobial activity of the phage at different temperatures

and pH. The bacteriophage presented complete stability at physiologic temperature and pH,

indicating that it could be used therapeutically. This study was then published in the Journal of

Basic Microbiology.
